Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_007078.3(LDB3):c.718G>C (p.Asp240His), citing Ambry Variant Classification Scheme 2023: The p.D240H variant (also known as c.718G>C), located in coding exon 5 of the LDB3 gene, results from a G to C substitution at nucleotide position 718. The aspartic acid at codon 240 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
