Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.7186G>A (p.Asp2396Asn), citing Ambry Variant Classification Scheme 2023: The p.D2396N variant (also known as c.7186G>A), located in coding exon 44 of the DNAH11 gene, results from a G to A substitution at nucleotide position 7186. The aspartic acid at codon 2396 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
