Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.7153C>A (p.Leu2385Ile), citing Ambry Variant Classification Scheme 2023: The p.L2385I variant (also known as c.7153C>A), located in coding exon 57 of the FBN2 gene, results from a C to A substitution at nucleotide position 7153. The leucine at codon 2385 is replaced by isoleucine, an amino acid with highly similar properties, and is located in the TGFBP #07 domain. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
