Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001042492.3(NF1):c.7214T>G (p.Ile2405Ser), citing Ambry Variant Classification Scheme 2023: The p.I2384S variant (also known as c.7151T>G), located in coding exon 48 of the NF1 gene, results from a T to G substitution at nucleotide position 7151. The isoleucine at codon 2384 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
