Uncertain significance — the classification assigned by Ambry Genetics to NM_006904.7(PRKDC):c.7071G>T (p.Glu2357Asp), citing Ambry Variant Classification Scheme 2023: The p.E2357D variant (also known as c.7071G>T), located in coding exon 53 of the PRKDC gene, results from a G to T substitution at nucleotide position 7071. The glutamic acid at codon 2357 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
