Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000039.3(APOA1):c.703C>G (p.Leu235Val), citing Ambry Variant Classification Scheme 2023: The p.L235V variant (also known as c.703C>G), located in coding exon 3 of the APOA1 gene, results from a C to G substitution at nucleotide position 703. The leucine at codon 235 is replaced by valine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, valine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
