Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001277115.2(DNAH11):c.12362A>C (p.Tyr4121Ser), citing Ambry Variant Classification Scheme 2023: The p.Y4121S variant (also known as c.12362A>C), located in coding exon 75 of the DNAH11 gene, results from an A to C substitution at nucleotide position 12362. The tyrosine at codon 4121 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
