NM_018979.4(WNK1):c.6261T>G (p.Ile2087Met) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I2339M variant (also known as c.7017T>G), located in coding exon 25 of the WNK1 gene, results from a T to G substitution at nucleotide position 7017. The isoleucine at codon 2339 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
