Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.6C>G (p.Asp2Glu), citing Ambry Variant Classification Scheme 2023: The p.D2E variant (also known as c.6C>G), located in coding exon 1 of the BUB1 gene, results from a C to G substitution at nucleotide position 6. The aspartic acid at codon 2 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
