Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_004387.4(NKX2-5):c.697G>C (p.Asp233His), citing Ambry Variant Classification Scheme 2023: The p.D233H variant (also known as c.697G>C), located in coding exon 2 of the NKX2-5 gene, results from a G to C substitution at nucleotide position 697. The aspartic acid at codon 233 is replaced by histidine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
