Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000400.4(ERCC2):c.1234A>G (p.Lys412Glu), citing Ambry Variant Classification Scheme 2023: The p.K412E variant (also known as c.1234A>G), located in coding exon 12 of the ERCC2 gene, results from an A to G substitution at nucleotide position 1234. The lysine at codon 412 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
