Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000890.5(KCNJ5):c.67A>G (p.Lys23Glu), citing Ambry Variant Classification Scheme 2023: The p.K23E variant (also known as c.67A>G), located in coding exon 1 of the KCNJ5 gene, results from an A to G substitution at nucleotide position 67. The lysine at codon 23 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
