Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_005120.3(MED12):c.679G>C (p.Glu227Gln), citing Ambry Variant Classification Scheme 2023: The p.E227Q variant (also known as c.679G>C), located in coding exon 5 of the MED12 gene, results from a G to C substitution at nucleotide position 679. The glutamic acid at codon 227 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
