Uncertain significance for Maturity-onset diabetes of the young — the classification assigned by Ambry Genetics to NM_000209.4(PDX1):c.677A>C (p.Asp226Ala), citing Ambry Variant Classification Scheme 2023: The p.D226A variant (also known as c.677A>C), located in coding exon 2 of the PDX1 gene, results from an A to C substitution at nucleotide position 677. The aspartic acid at codon 226 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
