Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.6745C>G (p.Gln2249Glu), citing Ambry Variant Classification Scheme 2023: The p.Q2249E variant (also known as c.6745C>G), located in coding exon 29 of the AKAP9 gene, results from a C to G substitution at nucleotide position 6745. The glutamine at codon 2249 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
