NM_000143.4(FH):c.667A>G (p.Lys223Glu) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K223E variant (also known as c.667A>G), located in coding exon 5 of the FH gene, results from an A to G substitution at nucleotide position 667. The lysine at codon 223 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
