NM_001277115.2(DNAH11):c.6676G>C (p.Asp2226His) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D2226H variant (also known as c.6676G>C), located in coding exon 40 of the DNAH11 gene, results from a G to C substitution at nucleotide position 6676. The aspartic acid at codon 2226 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
