This variant lies in the CLCN1 gene (transcript NM_000083.3) at coding-DNA position 566, where C is replaced by T; at the protein level this means replaces serine at residue 189 with phenylalanine — a missense variant. Submitter rationale: Variant summary: CLCN1 c.566C>T (p.Ser189Phe) results in a non-conservative amino acid change in the encoded protein sequence. Five of five in-silico tools predict a damaging effect of the variant on protein function. The variant was absent in 251486 control chromosomes. c.566C>T has been reported in the literature in individuals affected with Myotonia congenita (Dupre_2009). These data indicate that the variant may be associated with disease. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
