Submitter rationale: The p.D222Y variant (also known as c.664G>T), located in coding exon 4 of the MSH6 gene, results from a G to T substitution at nucleotide position 664. The aspartic acid at codon 222 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is well conserved in available vertebrate species.
