NM_000719.7(CACNA1C):c.664A>T (p.Asn222Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N222Y variant (also known as c.664A>T), located in coding exon 5 of the CACNA1C gene, results from an A to T substitution at nucleotide position 664. The asparagine at codon 222 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
