NM_005751.5(AKAP9):c.6637G>C (p.Glu2213Gln) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E2213Q variant (also known as c.6637G>C), located in coding exon 29 of the AKAP9 gene, results from a G to C substitution at nucleotide position 6637. The glutamic acid at codon 2213 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
