Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000059.4(BRCA2):c.6592G>C (p.Glu2198Gln), citing Ambry Variant Classification Scheme 2023: The p.E2198Q variant (also known as c.6592G>C), located in coding exon 10 of the BRCA2 gene, results from a G to C substitution at nucleotide position 6592. The glutamic acid at codon 2198 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
