NM_004415.4(DSP):c.6583C>T (p.His2195Tyr) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H2195Y variant (also known as c.6583C>T), located in coding exon 24 of the DSP gene, results from a C to T substitution at nucleotide position 6583. The histidine at codon 2195 is replaced by tyrosine, an amino acid with similar properties. This variant was detected in a cardiomyopathy genetic testing cohort; however, clinical details were limited, and additional cardiac variants were detected (van Lint FHM et al. Neth Heart J, 2019 Jun;27:304-309). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
