NM_018975.4(TERF2IP):c.646G>T (p.Asp216Tyr) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D216Y variant (also known as c.646G>T), located in coding exon 1 of the TERF2IP gene, results from a G to T substitution at nucleotide position 646. The aspartic acid at codon 216 is replaced by tyrosine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
