Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000784.4(CYP27A1):c.642G>T (p.Leu214Phe), citing Ambry Variant Classification Scheme 2023: The p.L214F variant (also known as c.642G>T), located in coding exon 3 of the CYP27A1 gene, results from a G to T substitution at nucleotide position 642. The leucine at codon 214 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
