NM_000890.5(KCNJ5):c.637G>A (p.Glu213Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E213K variant (also known as c.637G>A), located in coding exon 1 of the KCNJ5 gene, results from a G to A substitution at nucleotide position 637. The glutamic acid at codon 213 is replaced by lysine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
