NM_000527.5(LDLR):c.635_643dup (p.Trp214_Arg215insProSerTrp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The c.635_643dupCCAGCTGGC variant, located in coding exon 4 of the LDLR gene, results from an in-frame duplication of nine nucleotides, CCAGCTGGC, between nucleotide positions 635 and 643. This result in the insertion of three extra amino acid residues, proline, serine and tryptophan, between codons 214 and 215. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. Since supporting evidence is limited at this time, the clinical significance of this variant remains unclear.
