NM_000138.5(FBN1):c.6322C>G (p.Arg2108Gly) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R2108G variant (also known as c.6322C>G), located in coding exon 51 of the FBN1 gene, results from a C to G substitution at nucleotide position 6322. The arginine at codon 2108 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
