Uncertain significance — the classification assigned by Ambry Genetics to NM_002907.4(RECQL):c.631A>G (p.Arg211Gly), citing Ambry Variant Classification Scheme 2023: The p.R211G variant (also known as c.631A>G), located in coding exon 5 of the RECQL gene, results from an A to G substitution at nucleotide position 631. The arginine at codon 211 is replaced by glycine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
