Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_017617.5(NOTCH1):c.6314T>C (p.Met2105Thr), citing Ambry Variant Classification Scheme 2023: The p.M2105T variant (also known as c.6314T>C), located in coding exon 34 of the NOTCH1 gene, results from a T to C substitution at nucleotide position 6314. The methionine at codon 2105 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
