NM_004655.4(AXIN2):c.625T>A (p.Tyr209Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y209N variant (also known as c.625T>A), located in coding exon 1 of the AXIN2 gene, results from a T to A substitution at nucleotide position 625. The tyrosine at codon 209 is replaced by asparagine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
