Uncertain significance — the classification assigned by Ambry Genetics to NM_003579.4(RAD54L):c.621C>G (p.Cys207Trp), citing Ambry Variant Classification Scheme 2023: The p.C207W variant (also known as c.621C>G), located in coding exon 7 of the RAD54L gene, results from a C to G substitution at nucleotide position 621. The cysteine at codon 207 is replaced by tryptophan, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
