NM_006231.4(POLE):c.6204G>C (p.Gln2068His) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q2068H variant (also known as c.6204G>C), located in coding exon 45 of the POLE gene, results from a G to C substitution at nucleotide position 6204. The glutamine at codon 2068 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
