NM_000059.4(BRCA2):c.10198A>T (p.Ser3400Cys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S3400C variant (also known as c.10198A>T), located in coding exon 26 of the BRCA2 gene, results from an A to T substitution at nucleotide position 10198. The serine at codon 3400 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
