Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_017802.4(DNAAF5):c.619T>C (p.Ser207Pro), citing Ambry Variant Classification Scheme 2023: The p.S207P variant (also known as c.619T>C), located in coding exon 2 of the DNAAF5 gene, results from a T to C substitution at nucleotide position 619. The serine at codon 207 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
