Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001378454.1(ALMS1):c.12179A>G (p.Lys4060Arg), citing Ambry Variant Classification Scheme 2023: The p.K4061R variant (also known as c.12182A>G), located in coding exon 20 of the ALMS1 gene, results from an A to G substitution at nucleotide position 12182. The lysine at codon 4061 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and arginine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
