NM_001277115.2(DNAH11):c.6183T>A (p.Asp2061Glu) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D2061E variant (also known as c.6183T>A), located in coding exon 37 of the DNAH11 gene, results from a T to A substitution at nucleotide position 6183. The aspartic acid at codon 2061 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
