Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001267550.2(TTN):c.89030G>A (p.Ser29677Asn), citing Ambry Variant Classification Scheme 2023: The p.S20612N variant (also known as c.61835G>A), located in coding exon 160 of the TTN gene, results from a G to A substitution at nucleotide position 61835. The serine at codon 20612 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
