This variant lies in the TGFBR1 gene (transcript NM_004612.4) at coding-DNA position 615, where T is replaced by G; at the protein level this means replaces isoleucine at residue 205 with methionine — a missense variant. Submitter rationale: The p.I205M variant (also known as c.615T>G), located in coding exon 4 of the TGFBR1 gene, results from a T to G substitution at nucleotide position 615. The isoleucine at codon 205 is replaced by methionine, an amino acid with highly similar properties, and is in the protein kinase domain. This alteration was reported in a study of clinical genetic testing; however, clinical details were limited (Pepin MG et al. Genet. Med., 2016 Jan;18:20-4). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
