Pathogenic for Diamond-Blackfan anemia — the classification assigned by Ambry Genetics to NM_000969.5(RPL5):c.60del (p.Phe20fs), citing Ambry Variant Classification Scheme 2023: The c.60delT pathogenic mutation, located in coding exon 2 of the RPL5 gene, results from a deletion of one nucleotide at nucleotide position 60, causing a translational frameshift with a predicted alternate stop codon (p.F20Lfs*18). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
