Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001034853.2(RPGR):c.607G>C (p.Ala203Pro), citing Ambry Variant Classification Scheme 2023: The p.A203P variant (also known as c.607G>C), located in coding exon 6 of the RPGR gene, results from a G to C substitution at nucleotide position 607. The alanine at codon 203 is replaced by proline, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
