Uncertain significance — the classification assigned by Ambry Genetics to NM_004336.5(BUB1):c.607A>G (p.Lys203Glu), citing Ambry Variant Classification Scheme 2023: The p.K203E variant (also known as c.607A>G), located in coding exon 7 of the BUB1 gene, results from an A to G substitution at nucleotide position 607. The lysine at codon 203 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
