Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000335.5(SCN5A):c.6008T>G (p.Phe2003Cys), citing Ambry Variant Classification Scheme 2023: The p.F2004C variant (also known as c.6011T>G), located in coding exon 27 of the SCN5A gene, results from a T to G substitution at nucleotide position 6011. The phenylalanine at codon 2004 is replaced by cysteine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
