Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004655.4(AXIN2):c.5G>A (p.Ser2Asn), citing Ambry Variant Classification Scheme 2023: The p.S2N variant (also known as c.5G>A), located in coding exon 1 of the AXIN2 gene, results from a G to A substitution at nucleotide position 5. The serine at codon 2 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this alteration remains unclear.
