Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000535.7(PMS2):c.-5C>G, citing Ambry Variant Classification Scheme 2023: The c.-5C>G variant is located in the 5' untranslated region (5&rsquo; UTR) of the PMS2 gene. This variant results from a C to G substitution 5 bases upstream from the first translated codon. Based on nucleotide sequence alignment, this position is poorly conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
