Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000546.6(TP53):c.-5C>G, citing Ambry Variant Classification Scheme 2023: The c.-5C>G variant is located in the 5' untranslated region (5’ UTR) of the TP53 gene. This variant results from a C to G substitution 5 bases upstream from the first translated codon. This nucleotide position is well conserved in available vertebrate species. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
