Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_000384.3(APOB):c.5A>C (p.Asp2Ala), citing Ambry Variant Classification Scheme 2023: The p.D2A variant (also known as c.5A>C), located in coding exon 1 of the APOB gene, results from an A to C substitution at nucleotide position 5. The aspartic acid at codon 2 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
