Uncertain significance — the classification assigned by Ambry Genetics to NM_003579.4(RAD54L):c.598C>T (p.Leu200Phe), citing Ambry Variant Classification Scheme 2023: The p.L200F variant (also known as c.598C>T), located in coding exon 7 of the RAD54L gene, results from a C to T substitution at nucleotide position 598. The leucine at codon 200 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
