Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_005751.5(AKAP9):c.5947A>C (p.Met1983Leu), citing Ambry Variant Classification Scheme 2023: The p.M1983L variant (also known as c.5947A>C), located in coding exon 24 of the AKAP9 gene, results from an A to C substitution at nucleotide position 5947. The methionine at codon 1983 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species; however, leucine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
