Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001753.5(CAV1):c.58G>A (p.Glu20Lys), citing Ambry Variant Classification Scheme 2023: The p.E20K variant (also known as c.58G>A), located in coding exon 2 of the CAV1 gene, results from a G to A substitution at nucleotide position 58. The glutamic acid at codon 20 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
